生脉散对2型糖尿病大鼠心肌细胞的保护作用
Protective effect of Shengmaisan on myocardial cells of T2DM rats

Abstract
Objective To investigate the protective effect of Shengmaisan(SMS)on myocardial cells of type 2 diabetic(T2DM)rats.Methods Fifty sexually mature male SD rats were randomly divided into five groups according to the random num‑ber table method:control group,T2DM group,Shengmaisan intervention group[0.9 g/(kg·d),1.8 g/(kg·d),3.6 g/(kg·d)],with 10 rats in each group.After successful establishment of diabetes in the rats,the T2DM group was given normal saline by gavage,and the Shengmaisan groups were given different concentrations of Shengmaisan[0.9 g/(kg·d),1.8 g/(kg·d),3.6 g/(kg·d)]by gavage for 4W.After fasting for 8h after the last gavage,the weight of rats was weighed after isoflurane inhalation anesthesia,and the abdominal aorta blood was drawn by laparotomy to detect the relevant indicators(FBG,2‑hour postprandial blood glucose(2hPBG),fasting plasma insulin(FINS),insulin resistance index(HOMA‑IR),total cholesterol(TC),total triglycerides(TG),low‑density lipoprotein cholesterol(LDL‑C),high‑density lipoprotein cholesterol(HDL‑C)).The heart was quickly re‑moved and put into paraformaldehyde(HE staining,Masson staining,PAS staining,TUNEL staining,immunofluorescence stain‑ing(Bax,bcl‑2))and dry ice zone detection(Bax,bcl‑2,and activity of caspase‑9/3).Results Compared with the control group,FBG,2hpbg,FINS,HOMA‑IR,TG,TC,and LDL‑C increased in T2DM group(P<0.05),and decreased after intervention with Shengmaisan(P<0.05);HDL‑C in T2DM group decreased(P<0.05),and increased after the intervention of Shengmaisan(P<0.05);the degree of myocardial injury(HE staining),fibrosis(Masson staining),glycogen deposition(PAS staining),and apop‑tosis(TUNEL staining and activity of caspase‑9/3)in T2DM group increased(P<0.05),and decreased after the intervention of Shengmaisan(P<0.05);the expression of Bax(immunofluorescence and WB)in T2DM group increased(P<0.05),and de‑creased after the intervention of Shengmaisan(P<0.05)while the expression of Bcl‑2(immunofluorescence and WB)de‑creased(P<0.05),and increased after the intervention of Shengmaisan(P<0.05).Conclusions Shengmaisan can pro‑tect myocardial cells by improving apoptosis of myocardial tissue in T2DM rats.关键词
