鸭精液冷冻程序优化及抗氧化效果研究
Optimization of Cryopreservation Protocols and Antioxidant Efficacy in Duck Semen

Abstract
As a pivotal technique for genetic resource preservation and artificial breeding in waterfowl,sperm cryopreservation faces challenges due to the unique morphological and biochemical characteris-tics of avian spermatozoa,which render them highly susceptible to oxidative stress-induced motility decline during freezing.Using the Jinding duck as a model,this study systematically evaluated theef-fects of four distinct cryopreservation protocols(P1-P4)on sperm quality and oxidative stress re-sponses.A programmable freezer was used to execute multi-stage temperature modulation strategies,with sperm parameters quantified via automated sperm analysis and oxidative stress biomarkers as-sessed.The results demonstrated that Protocol P4-featuring an initial slow cooling phase(7℃/min to-35 ℃)followed by rapid freezing(60 ℃/min to-140 ℃)—achieved superior post-thaw out-comes:sperm viability reached 71.41%,mortality decreased to 28.59%,and progressive motility maintained at 46.99%,all significantly better than those in other protocols.In contrast,Protocol P2 exhibited the poorest performance(viability:34.28%;mortality:65.72%;motility:22.69%).An-tioxidant profiling revealed that P4-maintained superoxide dismutase(SOD)activity at levels compa-rable to fresh semen,while catalase(CAT)activity was significantly higher than that in P2.Notably,malondialdehyde(MDA)levels in P4 showed no significant elevation,whereas P2 demonstrated marked reductions in SOD,CAT,and total antioxidant capacity(T-AOC),along with significant MDA accumulation.Mechanistically,the biphasic cooling strategy of P4 optimized the balance between cellu-lar dehydration and ice crystal inhibition while minimizing the duration of hyperosmotic exposure,thereby effectively reducing reactive oxygen species(ROS)generation and oxidative damage.Con-versely,P2's multi-stage gradual cooling prolonged hypothermic stress,exacerbating mitochondrial dysfunction and oxidative stress amplification.This study demonstrates that cryopreservation proto-cols directly affect duck sperm quality by modulating oxidative stress,providing theoretical support for the standardization of avian semen cryopreservation technologies in genetic conservation programs.关键词
