基于线粒体D-loop区多样性的四平叶赫苍鹭种群动态研究
Population Dynamics of Grey Herons in Yehe,Siping Based on Diversity of Mitochondrial D-loop Region

Abstract
To explore the population dynamics of the breeding population of grey herons,209 allantoic membrane samples of grey heron eggshells were collected during 2021 to 2024 in the Yehe Scenic Are-a,Siping,Jilin Province.Genomic DNA was extracted,and the mitochondrial DNA D-loop region se-quences were amplified and sequenced.The base composition and content,variable sites,number of haplotypes,genetic diversity,and genetic differentiation coefficient of the 209 grey heron mtDNA D-loop region sequences were analyzed to assess the degree of sequence variation,and a haplotype net-work diagram was constructed.The results showed that the mitochondrial D-loop region sequences were successfully obtained for all samples.Analysis of base composition revealed that the average per-centages of T,C,A,and G were 27.99%,26.54%,25.63%,and 19.84%,respectively.A total of 55 mutation sites were identified,forming 33 haplotypes.Genetic diversity analysis showed that the hap-lotype diversity(Hd),nucleotide diversity(Pi),and average number of nucleotide differences(K)were 0.989,0.007 16 and 5.373,respectively,suggesting high genetic diversity(Hd≥0.5,Pi 0.005).The genetic differentiation analysis showed that the overall Fst value was 0.0293(Fst<0.05),indicating a slight genetic differentiation,although the result was statistically significant(P<0.05).As migratory birds,grey herons may experience gene flow during migration.The AMOVA analysis showed that among the overall genetic variation,the variation within the population accounted for 97.07%,which was significantly higher than that between populations(2.93%).The results of the neutral test showed that Tajima's D was-2.245 79 and Fu's Fs was-31.662,strongly sugges-ting a recent population expansion.Field observations also indicated a year-by-year increase in grey heron numbers in the Yehe Scenic Area.It is speculated that grey herons have strong environmental adaptability and development of a larger and more stable population.关键词
