LncRNA NEAT1 up-regulate CKIP-1 by inhibiting miR-98-5p to affect collagen secretion and apoptosis of pelvic floor supportive fibroblasts

Abstract
Objective:To investigate whether LncRNA NEAT1 up-regulates CKIP-1 ex-pression by inhibiting miR-98-5p and thereby affects collagen secretion and apoptosis in fibro-blasts of pelvic floor supporting tissue.Methods:Vaginal wall tissue collected from patients who had surgery for pelvic organ prolapse were classified as the POP group,and vaginal wall tissue from patients who hadsterectomy for other benign gynecological diseases were classified as con-trols.The expressions of LncRNA NEAT1,miR-98-5p,and CKIP-1 were detected.Culture of the vaginal wall fibroblasts was conducted.Using the cell-based transfection technique,CKIP-1 was knocked down,miR-98-5p was overexpressed,and LncRNA NEAT1 was knocked down.The ex-pressions of COL-Ⅰ,COL-Ⅲ,TGF-β1,Smad3,p-Smad3,Bax,and Bcl-2 were assessed by Western blot.Results:The expression of CKIP-1 in the POP group increased.After knockdown of CKIP-1,the expression of COL-Ⅰ,COL-Ⅲ,TGF-β1,Smad3,p-Smad3,and Bcl-2 were in-creased,while the expression of Bax was decreased.The expression of miR-98-5p in the POP group was decreased.After overexpression of miR-98-5p,the expressions of COL-Ⅰ,COL-Ⅲ,TGF-β1,Smad3,p-Smad3,and Bcl-2 were increased,while the expression of CKIP-1 and Bax were decreased.This result could be interfered with by CKIP-1.The expression of LncRNA NEAT1 in the POP group was increased.After knockdown of NEAT1,the expressions of COL-Ⅰ,COL-Ⅲ,TGF-β1,Smad3,p-Smad3,and Bcl-2 were increased,while the expressions of CK-IP-1 and Bax were decreased.This result was disturbed by miR-98-5p.Conclusion:LncRNA NEAT1 upregulated CKIP-1 expression by inhibiting miR-98-5p and then affected type Ⅰ and Ⅲ collagen secretion and apoptosis in fibroblasts of pelvic floor supporting tissues.关键词
