黄芪多糖调节Wnt/β-catenin信号通路对结直肠癌细胞5-氟尿嘧啶敏感性的影响
Effect of astragalus polysaccharide on sensitivity of colorectal cancer cells to 5-FU via regulating Wnt/β-catenin signaling pathway

Abstract
Objective It is to investigate the effect and possible mechanism of astragalus polysaccharide(AP)on the-sensitivity of colorectal cancer cells to 5-fluorouracil(5-FU)based on Wnt/β-catenin signaling pathway.Methods The half inhibitory concentrations(IC50)of AP and 5-FU to human colorectal cancer HCT116 cells were measured by MTT method,which were used as the medication concentrations for subsequent experiments.The HCT116 cells were taken and divided into blank control group,AP group,5-FU group,AP+5-FU group and AP+5-FU+Wnt/β-catenin signaling path-way agonist SKL2001 group.After 48 h of responding medicaiton intervention,the proliferation ability,survival rate,apop-tosis rate and autophagy of the cells were detected by cloning formation experiment,MTT experiment,flow cytometry and GFP-LC3 fluorescence plasmid transfection experiment,the mRNA expressions of Wnt3a and β-catenin in the cells were detected by RT-PCR,and the protein expressions of Wnt3a,β-catenin,Bcl-2,Bax,cleaved Caspase-3,Beclin1 and mi-crotubule-associated protein 1 light chain 3(LC3)were detected by Western blot.Results Compared with the blank control group,the number of HCT116 cell clones and the survival rates in AP group,5-FU group,AP+5-FU group,AP+5-FU+SKL2001 group were obviously decreased(all P<0.05),and the apoptosis rate and number of autophagosome were obvi-ously increased(all P<0.05);compared with AP group,5-FU group and AP+5-FU+SKL2001 group,the above effects regulating proliferation,survival,apoptosis and autophagy of the AP+5-FU group were enhanced,the differences were sig-nificant(all P<0.05).Compared with the blank control group,the relative mRNA expressions of Wnt3a,β-catenin,as well as the relative protein expressions of Wnt3a,β-catenin,Bcl-2 in the cells of AP group,AP+5-FU group and AP+5-FU+SKL2001 group were obviously decreased(all P<0.05),the relative protein expressions of Bax,cleaved Caspase-3,Beclin1,LC3Ⅰ,LC3Ⅱ and Bax/Bcl-2,LC3-Ⅱ/LC3-Ⅰ were obviously increased(all P<0.05).Compared with 5-FU group and AP+5-FU+SKL2001 group,the relative mRNA expressions of Wnt3a,β-catenin,as well as the relative pro-tein expressions of Wnt3a,β-catenin,Bcl-2 in the cells of AP+5-FU group were obviously decreased(all P<0.05),the relative protein expressions of Bax,cleaved Caspase-3,Beclin1,LC3Ⅰ,LC3Ⅱand Bax/Bcl-2,LC3-Ⅱ/LC3-Ⅰwere ob-viously increased(all P<0.05).Conclusion Astragalus polysaccharide may promote apoptosis and autophagy via inhibi-ting Wnt/β-catenin signaling pathway,thus to enhance the sensitivity of colorectal cancer cells to 5-FU.关键词
