摘要

Abstract

Objective It is to explore the effect of decoction for tonifying kidney and soothing liver on ovarian function and apoptosis of granulosa cells in rats with ovarian insufficiency based on nuclear factor erythroid 2 related factor 2(Nrf2)/antioxidant response element(ARE)pathway.Methods Sixty female SD rats were taken,10 rats of them were randomly se-lected as control group,and the other rats were given tripterygium glycosides by gavage to establish models of ovarian insuf-ficiency.The successfully modeled rats were randomly divided into model group,low-dose,medium-dose and high-dose groups of decoction for tonifying kidney and soothing liver,and western medicine group,with 10 rats in each group.The low-dose,medium-dose and high-dose groups of decoction for tonifying kidney and soothing liver were respectively adminis-tered decoction for tonifying kidney and soothing liver 1.9 g/kg,3.8 g/kg and 7.6 g/kg by gavage,and the western medi-cine group was administered Bardoxolone Methyl 30 mg/kg by gavage,the control group and model group were administered an equal volume of physiological saline by gavage,all once daily,continuously gavage for 15 days.The estrous cycles of rats in each group were recorded,the serum levels of gonadotropin-releasing hormone(GnRH),anti-Müllerian hormone(AMH),estradiol(E2),follicle-stimulating hormone(FSH)and luteinizing hormone(LH)were measured by ELISA,the ovarian tissue pathology was observed by HE staining,and the apoptosis rate of ovarian granulosa cells was determined by TUNEL staining method,the mRNA and protein expressions of Nrf2,Glutamate-cysteine ligase catalytic subunit(GCLC),NADPH quinone oxidoreductase 1(NQO1)and heme oxygenase-1(HO-1)in ovarian tissue were detected by RT-qPCR and Western blot methods.Results Compared with the control group,the estrous cycle of rats in the model group was significantly prolonged(P<0.05),and the serum levels of AMH and E2,as well as the relative mRNA and protein expressions of Nrf2,GCLC,NQO1,HO-1 in ovarian tissue were significantly decreased(all P<0.05),the serum levels of GnRH,FSH and LH,as well as the apoptosis rate of ovarian granulosa cells were significantly increased(all P<0.05).Compared with the model group,the estrous cycles of rats in each dose group of decoction for tonifying kidney and soothing liver and western medicine group were significantly shortened(all P<0.05),and the serum levels of AMH and E2,as well as the relative mRNA and protein expressions of Nrf2,GCLC,NQO1,HO-1 in ovarian tissue were significantly increased(all P<0.05),the serum levels of GnRH,FSH and LH,as well as the apoptosis rate of ovarian granulosa cells were significantly decreased(all P<0.05);the improvements of the above indicators in the medium-dose and high-dose groups of decoction for tonifying kidney and soothing liver and western medicine group were significantly better than those in the low-dose group(all P<0.05),the improvements in the high-dose group and western medicine group were significantly better than those in the medium-dose group(all P<0.05).Conclusion Decoction for tonifying kidney and soothing liver can significantly improve ovarian function,inhibit ovarian granulosa cell apoptosis in rats with ovarian insufficiency,and its mechanism may be related to regulation of Nrf2/ARE signaling pathway.
