摘要

Abstract

In this study,polyclonal antibodies to PPV VP2 were designed,prepared,and validated with porcine parvovirus(PPV)and eukaryotic plasmid pcDNA3.1(+)-VP2-Myc.PPV VP2 was then cloned into the prokaryotic expression vector pCold-TF,and VP2 pro-tein was induced by 1 mmol/L IPTG.Next,the purified VP2 protein was immunized against New Zealand white rabbits,and polyclonal anti-bodies were prepared and subjected to ELISA,Western blot;and IFA was used to verify their reactivity.The results of ELISA showed that the titer of the polyclonal antibody was as high as 1∶128 000.The results of Western blot and IFA showed that the antibody could specifical-ly recognize and bind to PPV,indicating that the recombinant VP2 protein had good immunogenicity and reactogenicity.This study is condu-cive to the establishment of a VP2 serological detection method and the functional study of VP2 protein.
