Structure and characteristics analysis of Persicaria capitata chloroplast genome

Abstract
[Objective]The high-quality whole-genome information of the chloroplast in the medicinal plant Persicaria capitata was explored,and its structure,sequence characteristics and phylogenetic relationship were clarified,which provided references for its molecular identifica-tion,genetic diversity research,germplasm resources protection,development and utilization.[Method]The chloroplast genome structure of P.capitata was studied by the Illumina Novaseq 6000 sequencing platform and bioinformatics software,and its sequence features,repetitive se-quences,genetic diversity and phylogenetic relationship were analyzed.[Result]The total length of the chloroplast genome of P.capitata was 158 817 bp.The lengths of the IRa and IRb were both 30 931 bp,and the lengths of the LSC and SSC were 84 049 and 12 906 bp,respec-tively.The total GC content of the chloroplast genome was 38.0%,and it contained 127 coding genes,including 82 protein-coding genes,37 tRNA genes and 8 rRNA genes.Among them,19 genes such as ndhBa,b,rrn16a and rrn23a had two copies,15 genes such aspetBb,petDband atpFb had one intron,and 3 genes(rps12a,c,clpP(and ycf3c)had two introns.P.capitata clustered with its congener P.chinensis into one clade with the support rate of 100%.[Conclusion]The high-quality chloroplast genome of P.capitata is successfully assembled,and the in-formation on the structure and sequence characteristics of its chloroplast genome is obtained,which lays the theoretical foundation for the i-dentification,evolution and phylogenetic research of medicinal plants in the genus Persicaria.关键词
