沙棘MBF1基因家族鉴定及铅胁迫下表达分析
Identification and Pb stress expression analysis of Hippophae rhamnoides MBF1 gene family

Abstract
[Objective]The study aimed to identifiy members of the MBF1 gene family in Hippophae rhamnoides at the whole-genome level,and analyze their gene expression patterns,thereby establishing a theoretical foundation for subsequent studies on the mechanisms through which the MBF1 gene family responds to heavy metal lead stress.[Method]Members of the H.rhamnoides MBF1 gene family were identified using bioinformatics methods,and their physicochemical properties,subcellular and chromosomal localizations,as well as cis-acting elements,were analyzed.Additionally,their expression patterns under Pb stress were examined through transcriptomic and RT-qPCR analyses.[Result]Six MBF1 gene family members were identified in H.rhamnoides,designated as HrMBF1-1 to HrMBF1-6;The HrMBF1 gene-encoded pro-teins consist of 110 to 144 amino acids,with molecular weights ranging from 12 441.43 to 16 002.43 Da.Their isoelectric points fall within the range of 9.91 to 10.18,indicating that these proteins were basic.The instability indiced range from 34.72 to 60.07,with HrMBF1-1,HrMBF1-3 and HrMBF1-4 being stable proteins,while HrMBF1-2,HrMBF1-5 and HrMBF1-6 were unstable.The aliphatic index were be-tween 79.15 and 89.55.The average hydropathicity coefficients ranged from-0.645 to-0.845,all of which were less than zero,indicating that all six HrMBF1 gene family members were hydrophilic proteins.The secondary structures of these proteins mainly included α-helices,β-sheets,turns and random coils.Chromosomal localization revealed that the six HrMBF1 genes were unevenly distributed across five chromo-somes.Phylogenetic analysis divides the HrMBF1 gene family into two groups:Group I comprises HrMBF1-1,HrMBF1-3 and HrMBF1-4,which were localized in the nucleus;Group Ⅱ included HrMBF1-2,HrMBF1-5 and HrMBF1-6,which were localized in the cytoplasm and predicted to be nuclear or cytoplasmic.Domain analysis indicates that,apart from HrMBF1-2,which contains only the aMBF1 domain,the re-maining HrMBF1 gene family members possesed both MBF1 and aMBF1 domains.The HrMBF1 genes contain seven conserved motifs.H.rh-amnoides MBF1 genes contained cis-acting elements associated with abiotic stresses such as heat,drought and growth;The duplication event analysis revealed that there were four pairs of H.rhamnoides MBF1 genes with duplicated segments.Interspecific collinearity relationships were observed between H.rhamnoides and both Arabidopsis and Populus trichocarpa;transcriptome and RT-qPCR validation revealed that,with increasing concentrations of Pb stress,the expression levels of all MBF1 gene family members,except for HrMBF1-2,exhibited a trend of initial increase followed by decrease and then another increase.[Conclusion]The MBF1 gene family members play an important role in re-sponding to Pb stress in shrub species.关键词
