摘要

Abstract

[Objective]To explore the phenotypic changes and physiological responses of Barringtonia racemosa seedlings leaves under natural advection cold stress,and provide reference for cold damage defense,cultivation,conservation and the promotion of B.racemosa.[Method]Three-year-old B.racemosa seedlings were used as materials,with seedlings without cold stress serving as the control.The effects of different degrees of natural cold injury(mild cold injury,moderate cold injury,severe cold injury)on leaf morphological changes and physiological in-dexes of B.racemosa seedlings were studied in Yulin during the introduction,and correlation analysis and principal component analysis were carried out.[Result]As the severity of the cold increased,the leaves of B.racemosa gradually exhibited browning,chlorosis,curling and leaf drop.In terms of leaf phenotype,compared with control,both leaf area and leaf length significantly decreased(P<0.05,the same as below);as the severity of the cold increased,leaf perimeter showed a trend of initially increasing and then decreasing,while leaf width and the leaf length/leaf width ratio fluctuated to varying degrees.In the leaf antioxidant system,compared with control,under cold environment,the activi-ty of peroxidase(POD)significantly increased;as the severity of the cold increased,while ascorbate peroxidase(APX)activity initially in-creased and then decreased.The content of malondialdehyde(MD A),a product of lipid peroxidation products,and proline(Pro),an osmotic regulator,both exhibited a trend of initially increasing and then decreasing.As the severity of the cold increased,the content of photosynthetic pigments in B.racemosa showed a trend of first increasing and then decreasing,the contents of chlorophyll a,chlorophyll b,total chlorophyll and carotenoid were the highest and the lowest in control under mild cold injury.Correlation analysis showed that APX activity was signifi-cantly positively correlated with chlorophyll a content and carotenoid content,leaf area was significantly positively correlated with leaf width,POD activity was extremely significantly positively correlated with chlorophyll b content(P<0.01).Principal component analysis showed that the cumulative contribution rate of principal component 1 and principal component 2 was 76.478%,and the photosynthesis of the leaves of the seedlings had adaptive changes,and formed a chain reaction with some physiological indicators.[Conclusion]B.racemosa can withstand mild and moderate cold stress.Under cold stress,B.racemosa enhances its cold tolerance by increasing APX activity,POD activity,Pro con-tent,and photosynthetic pigment levels.These indicators can serve as potential markers for evaluating the cold tolerance of B.racemosa.
