广西西番莲产区东亚西番莲病毒的发生情况及其遗传多样性分析
Survey and genetic diversity analysis of East Asian passiflora virus in passion fruit of Guangxi

Abstract
[Objective]The investigation of East Asian passiflora virus(EAPV)occurrence in passion fruit in Guangxi,along with the analysis of EAPV population genetic structure were studied,providing reference for elucidating the epidemiological patterns of EAPV in the field and formulating effective prevention and control strategies.[Method]A total of 187 passion fruit samples,suspected to be infected with virus,were collected from the primary production areas across 13 cities in Guangxi.These samples were subsequently identified using RT-PCR and nucle-otide sequence comparison analysis.Regionally representative EAPV positive samples were selected,and based on the complete coat protein(CP)gene sequence,sequence similarity,genetic diversity,genetic variation,population structure and other characteristics were analyzed.[Result]Through RT-PCR analysis,132 samples tested positive for EAPV,yielding a detection rate of 70.6%.From the positive samples,23 EAPV coat protein gene sequences were obtained,which were subsequently uploaded to GenBank to obtain accession numbers.The nucleotide and amino acid sequence identities of CP gene from the 23 EAPV isolates ranged from 80.9%to 100.0%and from 83.4%to 100.0%re-spectively.The phylogenetic tree revealed that 23 EAPV isolates belonged to the AO strain(8 isolates)and the IB strain(15 isolates).Nu-cleotides 87-95 in CP protein coding region of IB strain were 9 more than those of AO strain.The results of genetic difference analysis showed that the genetic differentiation between the two EAPV strains was significant.The results of gene exchange showed that the frequency of gene exchange between the two strains was low,and genetic drift was easy to occur between the two strains.The results of genetic diversity analysis showed that the gene evolution of AO and IB lines was mainly affected by negative selection pressure,and the two strains were in the trend of expansion.[Conclusion]EAPV is widespread in the main growing areas of passion fruit in Guangxi.The 23 EAPV isolates obtained in this study contain AO strain and IB strain.Genetic drift and negative selection of genes are important evolution mechanisms for EAPV iso-lates.关键词
