摘要

Abstract

[Objective]The species of pathogen causing soft rot of Amorphphallus konjac seed tubers are identified in Weining county,Bijie re-gion,Guizhou province,and the effective bactericides against the pathogen of A.konjac soft rot were screened to provide a theoretical basis for control of A.konjac soft rot.[Method]The pathogenicity of the purified pathogen strains isolated from the diseased A.konjac seed tubers was detected by Koch's postulate and the purified pathogen strains were identified by molecular biology and physiological biochemistry.The indoor toxicity of 10 bactericides including 18%Chunleimycin.copper rosin acid SC,25%Brothermothingonil ME and 0.3%Tetramycin AS,etc.against the pathogen causing A.konjac soft rot was determined by the inhibition zone method to screen the bactericides with a significant anti-bacterial effect.[Result]The physiological and biochemical characteristics of the isolated pathogenic strain Akst2-3 causing soft rot of A.kon-jac seed tubers were consistent with Raoultella sp.and the homology between strain Akst2-3 and R.terrigena JCM 1687 reached 99.79%.The phylogenetic tree revealed that strain Akst2-3 and R.terrigena were located on the same branch.Therefore,strain Akst2-3 could be iden-tified as R.terrigena.The indoor toxicity measurement indicated that five bactericides of 18%Chunleimycin·copper rosin acid SC,80%Ethylicin EC,25%Brothermothingonil ME,0.3%Tetramycin AS and 5%Garlicin ME had the significant inhibitory effect against the patho-gen causing soft rot of A.konjac seed tubers.The EC50 of 18%Chunleimycin·copper rosin acid SC,80%Ethylicin EC,25%Brothermothin-gonil ME,0.3%Tetramycin AS and 5%Garlicin ME was 0.5282,1.3023,1.6882,2.0886 and 13.8071 mg/L respectively.[Conclusion]The R.terrigena is a new pathogen causing soft rot of A.konjac seed tubers.18%Chunleimycin·copper rosin acid SC and 80%Ethylicin EC can be selected as the preferred bactericides for controlling the native R.terrigena causing soft rot of A.konjac seed tubers.25%Bromothalo-nil ME and 0.3%Tetramycin AS can be used as the alternative bactericides in control of A.konjac soft rot.Four bactericides should be alter-nately and mixedly used to prevent the resistance risk of a single bactericide in A.konjac production.
