侵染枣树的枣长线病毒1鉴定及基因组序列分析
Identification and analysis of genomic sequence of jujube closterovirus 1 infecting jujube

Abstract
[Objective]The paper aimed to investigate the prevalence of Jujube Closterovirus 1(JCV1)in Dalate Banner,Ordos City,Inner Mongolia Autonomous Region,and to analyze the genetic evolutionary relationships among JCV1 isolates from different regions,providing a theoretical basis for the development of detection methods and scientific control strategies for JCV1.[Method]The leaves of jujube trees sus-pected of viral infection were taken as experimental materials,high-throughput sequencing was performed on jujube leaf samples using NGS sequencing technology,and the occurrence of JCV1 was detected by RT-PCR.The full-length genome sequence of JCV1 was obtained by com-bining RT-PCR with rapid amplification of cDNA ends(RACE)technology.Bioinformatics analysis software such as VectorNTI,MEGA11,SDTV1.2,RDP4,and Geneious Pro 4.8.4 were used to conduct phylogenetic analysis,sequence identity analysis,recombination analysis,and secondary structure prediction of the full-length genome sequences of JCV1 isolates.[Result]Through NGS sequencing,only JCV1 was detected in jujube samples exhibiting mosaic symptoms,indicating a close correlation between this virus and the mosaic symptoms of jujube trees in the sampling area.The detection results of RT-PCR showed that the detection rate of JCV1 in the jujube orchards of Dalate Banner,Ordos was 52%.The full-length genome sequence of the Inner Mongolia isolate 23IMZiJu1 of JCV1 obtained was 14 209 nt,encoding six open reading frames.The sequence identity analysis showed that the full-length genome nucleotide sequence identity of 23IM_ZiJu1 with the isolate from Xinjiang region(AKS15-20)and the amino acid sequence identity of its RdRp,HSP70,and CP genes were the highest,at 98.7%,99.1%,99.8%and 99.7%,respectively.The phylogenetic trees constructed based on the RdRp,HSP70,and CP genes clustered 23IM_ZiJu1 with two isolates from Xinjiang region,AKS15-20 and AKS15-17,in the same branch,and the genetic relationship between 23IM_ZiJu1 and AKS15-20 was the closest.Recombination analysis indicated that there was one significant recombination event in the genome of 23IM_ZiJu1.The differences in the secondary structure of proteins from JCV1 isolates from different regions were mainly reflected in the presence or absence and length of β-turns,β-sheets,and α-helices.[Conclusion]The study is the first to detect JCV1 isolates(23IM_ZiJu1)in Inner Mongolia region after Xinjiang region and obtain their genome sequences,as well as the evolutionary relationships with other known viruses in the same genus.The obtained genome sequence provides important reference for the research on genetic variation of JCV1 and the formulation of control strategies.关键词
