基于转录组测序分析3-磷酸甘油醛脱氢酶基因OsGAPC3对水稻生长发育的影响
Transcriptome sequencing analysis of effect of glyceraldehyde-3-phosphate dehydrogenase gene OsGAPC3 on growth and development of rice

Abstract
[Objective]The present paper analyzed the effects of the 3-phosphoglyceraldehyde dehydrogenase gene OsGAPC3 on rice growth,development,and quality based on transcriptome sequencing.The application potential of OsGAPC3 in rice molecular breeding was fully tapped into,thus providing theoretical basis for improving the quality and yield of rice.[Method]Transcriptome sequencing analysis of Os-gapc3 mutants and OsGAPC3 overexpressed transgenic materials were performed by high-throughput RNA-seq sequencing technology.Differ-entially expressed genes were screened and obtained,and GO and KEGG functional annotation and enrichment analysis on the differentially expressed genes were performed.[Result]241 genes were significantly upregulated and 485 genes were significantly downregulated in the Os-gapc3 homozygous mutant.827 genes were significantly upregulated and 1345 genes were significantly downregulated in OsGAPC3 transgenic positive plants.GO functional enrichment analysis showed that the differentially expressed genes in Osgapc3 mutants were mainly involved in biological processes such as flower development regulation,photoperiod flowering and defense response.The differentially expressed genes in OsGAPC3 overexpressing transgenic positive plants mainly participated in biological processes such as defense response,protein phosphoryla-tion and pollen recognition.The pollen iodine staining experiment showed that OsGAPC3 significantly affected the fertility of rice pollen.KEGG metabolic pathway enrichment analysis showed that the differentially expressed genes in Osgapc3 mutants were mainly enriched in bio-logical pathways such as metabolic pathways,biosynthesis of secondary metabolites,photosynthetic carbon fixation,oxidative phosphorylation and tyrosine metabolism.The differentially expressed genes in OsGAPC3 overexpressing transgenic plants were mainly enriched in biological pathways such as metabolic pathways,biosynthesis of secondary metabolites,plant hormone signal transduction,starch and sucrose metabolism and brassinosteroid biosynthesis.[Conclusion]Transcriptome sequencing analysis shows that OsGAPC3 is involved in various biological processes and metabolic pathways in rice,and regulates the carbon metabolism,growth and development and stress response of rice.This pro-vides important genetic resources for molecular design and breeding of high-quality,high-yield,stress resistant rice varieties.关键词
