荷花GRF基因家族鉴定及外源GA3和NAA处理表达分析
Identification and expression analysis of GRF gene family in Nelumbo nucifera exogenous GA3 and NAA treatments

Abstract
[Objective]The study aimed to investigate the influence of exogenous GA3(Gibberellic acid)and NAA(1-naphthlcetic acid)on the expression pattern of the lotus(Nelumbo nucifera Gaertn.)growth-regulating factor(GRF)gene family,thereby establishing a foundation for further research into the functions of the lotus NnGRF gene family.[Method]The NnGRF gene family of lotus at the whole genome level was identified and its basic characteristics were analyzed by using bioinformatics methods.'Tai Kong Lian'seedings were used as experimen-tal materials and treated with GA3 and NAA at concentrations of 1000 mg/L,10 mg/L,0.1 mg/L,1μg/L,0.01 μg/L,respectively,and the expression changes of the NnGRF gene family were analyzed under treatment.[Result]A total of 22 members of the lotus NnGRF gene family were identified,divided into 3 branches and 6 sub-branches.All NnGRF members included QLQ(Gln,Leu,Lln)and WRC(Try,Arg,Cys)domains,and the NnGRF promoter contained elements related to light,growth and development,abiotic stress,and plant hormone response;Compared with treatment CK,NnGRF21 showed a highly significant response to 1 μg/L GA3,increasing by more than 20 times of relative expression level;Under exogenous NAA treatment,the response of NnGRF14 to five concentrations of NAA was significantly up-regu-lated,and compared with treatment CK,it reached maximum value at 1 μg/L,with relative expression level increased by more than 200 times;NnGRF17 was significantly increased by more than 10 times under exogenous GA3(maximum at 10 mg/L)and NAA(maximum at 1μg/L)treatments.[Conclusion]The NnGRF14,NnGRF17 and NnGRF21 genes exhibit strong positive responses to exogenous GA3 and NA A,suggesting that they can serve as important candidate genes for investigating the regulatory mechanisms underlying lotus growth and de-velopment.The study reveals the GA3/NAA-specific expression patterns of NnGRF genes,which lays a critical foundation for further investi-gations into the gene function.关键词
