摘要

Abstract

[Objective]The study aimed to identify the members of transcription factor family of medicinal plant Angelica sinensis MYB(V-Mybavian Mylobastosis Viral Oncogene Homolog)and study its transcriptome expression pattern,screening MYB transcription factors that had potential in flavonoids synthesis.[Method]The study identified and analyzed the MYB family transcription based on the whole genome of A.sinensis,screened out candidate MYB family members that had potential to synthesize of flavonoids in A.sinensis.Bioinformatics methods were used to analyze the protein physical and chemical properties,gene structure,conserved motifs,conserved domains,phylogenetic development,chromosome localization,collinearity,cis-acting elements and transcriptome expression patterns of 153 MYB family transcription factors.[Re-sult]Through HMMER and BLAST screening of A.sinensis MYB transcription factors,a total of 153 MYB transcription factors were annotated in the whole genome of A.sinensis,21 of which were in the 1R-MYB subfamily,and the 2R-MYB subfamily had the largest number of mem-bers,with 130 members,excluding 3R-MYB and 2 MYB members in the 4R-MYB subfamily.The amino acid length of 153 transcription fac-tors ranged from 179 to 451 aa,and their relative molecular masses ranged from 20.0 to 50.1 kDa,all of which were hydrophilic proteins.Subcellular localization analysis showed that only one protein was localized in the nucleus and cytoplasm simultaneously,and the rest were lo-calized in the nucleus.According to the Arabidopsis MYB family classification,it was found that the transcription factor gene structures on the same branch were similar.Prediction of cis-acting elements revealed that elements related to growth and development,hormone regulation and stress were widely distributed in the MYB promoter region.Transcriptome differentially expressed gene analysis found that 121 transcription factors among 153 MYBs were differentially expressed in different parts of angelica roots and leaves.In addition,nine candidate MYB tran-scription factors that might be involved in flavonoid biosynthesis in A.sinensis were identified through phylogenetic analysis.The results of quantitative real-time PCR for these candidate genes were consistent with the transcription data.[Conclusion]Based on the whole genome da-ta of A.sinensis,153 MYB transcription factors are identified,including 21 members of 1R-MYB subfamily,130 members of 2R-MYB subfam-ily and 2 members of 4R-MYB subfamily,excluding 3R-MYB subfamily.RT-qPCR results show that AsMYB34,AsMYB79,AsMYB96 and AsMYB127 may be involved in the synthesis of flavonols in A.sinensis.
