半夏OFP基因家族的全基因组鉴定及表达模式分析
Genome-wide identification and expression pattern analysis of OFP gene family in Pinellia ternata

Abstract
[Objective]Ovate family proteins(OFPs)were a class of plant-specific proteins that played a pivotal role in regulating various as-pects of plant growth and development.The tuber of Pinellia ternate,which served as the principal medicinal component,had a substantial im-pact on the yield and quality of the plant.The objective of the study was to explore the structural features of OFP gene family members within P.ternate,and to assess their potential contributions to the tuber formation process.[Method]Based on the published chromosomal-level ge-nome assembly of P.ternate,bioinformatics methods were employed to identify the OFP gene family members in P.ternate.Following the i-dentification,a comprehensive analysis was carried out.This included examining gene structures,the physicochemical properties of the pro-teins,subcellular localization,conserved motifs,domains,tertiary structures,and promoter cis-acting elements.Phylogenetic,collinearity,and gene duplication and loss analysis were also undertaken,followed by an analysis of its expression patterns using transcriptomic data.[Result]A total of 25 PtOFP genes were identified.Analysis of the proteins' physicochemical properties and subcellular localization revealed that Pt-OFPs were unstable hydrophilic proteins primarily localized in the nucleus.Further examination of conserved motifs,structural domains,gene structure and protein tertiary structure indicated that PtOFPs exhibited relative conservation in both sequence and structure,suggesting poten-tial functional redundancy among some members.Phylogenetic,collinearity,and gene duplication and loss analysis revealed that the expansion of the P.ternata OFP gene family members mainly resulted from segmental duplication and tandem duplication,and they were evolutionarily closer to rice.Analysis of promoter cis-acting elements suggested that PtOFPs might be involved in response to biotic and abiotic stresses and regulate the growth and development of P.ternata through hormonal responses.The expression patterns of PtOFPs genes at different develop-mental stages of tubers and under exogenous brassinolide treatment conditions indicated that PtOFPs were very likely to be involved in the regulation of tuber developmental process through the brassinosteroid(BR)pathway,playing a role in maintaining tuber size.In particular,PtOFP9,PtOFP13 and PtOFP16 merited close attention.[Conclusion]The results of the study will contribute to revealing the involvement of PtOFPs in the tuber development process of P.ternata,thereby establishing a theoretical basis for future breeding,enhancing varieties,optimi-zing cultivation practices,and maximizing the utilization of P.ternata germplasm resources.关键词
