摘要

Abstract

To explore the serum metabolic profiles and rumen microbiota metabolome characteristics of Tibetan sheep at different altitudes,18 healthy 3.5-year-old female Tibetan sheep were selected from three altitudes(2 500 m,3 500 m and 4 500 m),with 6 each group,and the rumen microbial and serum metabolites were determined by LC-MS/MS tech-nology.The results showed that 1 394 rumen microbial metabolites were identified in the three altitude groups of Ti-betan sheep.Among them,fructosyl-lysine content was significantly higher in the 3 500 m group than in the other two altitude groups(P<0.05),while L-kynurenine content significantly decreased with increasing altitude(P<0.05).KEGG functional annotation showed that the differential metabolites between the 3 500 m and 4 500 m groups were mainly en-riched in amino acid,lipid and carbohydrate metabolism pathways.The metabolites in the 3 500 m group were up-regulated,mainly concentrated in the vitamin B6 metabolism pathway,while the 4 500 m group was enriched in pyrimi-dine metabolism.A total of 3 582 metabolites were identified in the serum of Tibetan sheep at different altitudes.The differential metabolites between the 3 500 m and 4 500 m groups were mainly enriched in amino acid metabolism,lipid metabolism and cytochrome P450-mediated xenobiotic metabolism pathways,and were correlated with microbial me-tabolites.Interaction analysis showed that 274 common metabolites were identified in rumen fluid and serum,and these common metabolites were mainly enriched in metabolism-related pathways.In conclusion,the microbial metabolites and serum metabolites of Tibetan sheep were co-enriched in pathways related to lipid metabolism,energy metabolism and immune metabolism,which may be involved in the adaptive regulation of Tibetan sheep to high-altitude environ-mental changes.
