摘要

Abstract

The aim of this study was to understand the transcriptome differences between X and Y spermatozoa in male Holstein cattle,screen the differential genes for validation,and provide candidate genes for the selection of X and Y sperm characteristic antigens.This study was based on transcriptome sequencing technology,using the Illumina plat-form to sequence X spermatozoa and Y spermatozoa of Holstein cattle with a purity of 95%.The results showed that 809 X sperm-specific expressed genes,1 939 Y sperm-specific expressed genes and 645 co-expressed genes were ob-tained.GO enrichment and KEGG pathway enrichment analyses were performed on the differential genes,and five pos-sible candidate genes were screened as segregating X and Y spermatozoa pathways(PRM1,PRAMEY,ITGA6,CD63,SCAMP1).RT-qPCR validation of the differential genes was performed,and it was found that the expression trend was in agreement with the results of the transcriptomes.By fluorescence in situ hybridisation,it was found that SCAMP1 mainly aggregated in the anterior part of the sperm head and tail,and PRAMEY mainly aggregated in the sperm tail.
