Effect of Feeding Chinese Herb Icariin on the Barrier Structure of Rumen Epithelium and Rumen Microbiota in Dairy Goats

Abstract
The aim of this study was to analyze the effect of feeding icariin(ICA)in the daily diet on the barrier struc-ture of rumen epithelium and rumen fluid microbiota in dairy goats,laying the foundation for the clinical application of ICA as a novel Chinese herbal feed additive.A total of 12 healthy goats with an average weight of approximately 38.95 kg±3.24 kg were randomly divided into two groups.The control group was fed the basal diet,and the ICA group was fed the basal diet supplemented with ICA(50 mg/kg·day)for 90 days,and the pre-test period was 10 days.Blood physi-ological and biochemical indicators were determined on days 0,10,20,40,60,and 80.After slaughtering the goats,the tissue sample of rumen was collected and the composition of the rumen fluid microbiota was analyzed by 16S rDNA se-quencing.The results showed that the white blood cell(WBC),lymphocyte(LYMPH)and mononuclear cell(MONON)counts in the ICA group showed an upward trend,but not statistically significant(P>0.05).After 60 days of ICA feeding,the red blood cell(RBC)and hematokrit(HCT)of dairy goats were significantly higher(P<0.05);while the mean corpuscular hemoglobin(MCH)and RDW were significantly lower in the ICA group compared to control group(P<0.05).Compared with the control group,serum glutamyl-aminotransferase(AST),alkaline phosphatase(ALP)and total bile acid(TBA)were significantly higher in the ICA group(P<0.05).Feeding ICA to dairy goats had no significant effect on the height and width of rumen papillae(P>0.05),but it could increase the thickness of the rumen wall,and it could sig-nificantly upregulate the expression levels of Occludin,ZO-1,Claudin-1,Claudin-4 and Claudin-7 genes in the rumen epi-thelium.16S rDNA sequencing analysis showed that a total of 1 350 OTUs(operational taxonomic units)were generated in the control group,of which 879 were unique.And the ICA group produced 2 605 OTUs,of which 2 143 were unique.Compared to the control group,the ICA group had a higher Shannon index,Chao index and ACE index.At the phylum level,the dominant phyla were Firmicutes,Bacteroidota,and Actinobacteriota for 2 groups.The relative abundance of Bacteroidota in the ICA group was significantly higher,while Actinobacteriota was lower compared to the control group(P<0.05).At the genus level,the relative abundance of Olsenella was significantly lower in the ICA group,while the abun-dance of Rikenellaceae_RC9_gut_group and Prevotella was higher than that in the control group(P<0.05).Prediction of microbial flora function by PICRUSt2 showed that the abundance of microbial communities related to carbohydrate me-tabolism,amino acid metabolism,terpenoid and polyketide metabolism of dairy goats in the experimental group was sig-nificantly lower than that in the control group(P<0.05).In summary,the addition of ICA to the daily diet of goats has a positive effect on strengthening immune ability and enhance the barrier structure of the rumen epithelium,and promote an increase of the relative abundances for beneficial bacteria.关键词
