摘要

Abstract

Objective To investigate the effects of phenyl-ethyl isothiocyanate(PEITC)on human colon cancer HCT116 cells and explore its molecular mechanism.Methods HCT116 cells were divided into control group and PEITC experimental-L,-M and-H dose groups,and SMAC mimetics,si-SMAC,and combination groups were also set up.The control group was treated with complete medium;the experimental-L,-M and-H groups were treated with 10,20,50 μmol·L-1 PEITC;the SMAC mimetics group and the si-SMAC group were transfected with second mitochondria-derived activator of caspases(SMAC)mimetics and SMAC synthetic double-stranded interference RNA(si-SMAC),respectively;the cell in combination group were treatment with si-SMAC for 24 h and then cultured with 20 μmol·L-1 PEITC.The cell proliferation was measured by cell counting rit8;cell clone formation and flow cytometry was used to detect the clone formation ability and apoptosis rate of HCT116 cells.The protein expressions of B-cell lymphoma-2(Bcl-2),caspase-3,caspase-8 and SMAC were detected by Western blot.Results After cultivating cells with 0-200 μmol·L-1 PEITC,the activity of HCT116 cells was significantly reduced and the IC50 value was 42.18μmol·L-1.Subsequently,in the control group and the experimental-L,-M and-H group,the number of colonies formed was 415.75±26.22,247.13±21.11,198.13±31.60 and 169.63±26.82,respectively;the early apoptosis rates were(3.13±1.01)%,(25.49±1.60)%,(34.29±1.77)%and(24.95±0.65)%;the protein relative expression levels of SMAC were 0.95±0.13,1.49±0.15,2.78±0.16 and 1.91±0.19;the protein relative expression levels of caspase-3 were 1.06±0.06,1.21±0.15,2.10±0.25 and 1.58±0.23;the protein relative expression levels of Bcl-2 were 0.97±0.05,0.67±0.05,0.29±0.08 and 0.59±0.06,respectively;compared with the control group,the above indicators of the experimental-L,-M and-H group were all significantly different(all P<0.01).The relative expression levels of caspase-3 in si-SMAC group and combination group were 0.57±0.06 and 1.65±0.09,the relative expression levels of Bcl-2 were 1.35±0.06 and 0.39±0.16,and the differences were statistically significant(all P<0.05).Conclusion PEITC can activate caspase-3 apoptosis pathway in HCT116 cells by up-regulating SMAC expression,thereby promoting cell apoptosis.
