Abstract
Objective To investigate the effects of inhaled aildenafil citrate liposomes on pulmonary artery pressure and pulmonary vascular remodeling in monocrotaline(MCT)-induced pulmonary arterial hypertension(PAH)rats.Methods PAH model was induced by MCT in rats.The rats were randomly divided into normal group(intraperitoneal injection of equal volume of 0.9%NaCl),model group(construction PAH rat model was constructed,and rats were intraperitoneally injected with 50 mg·kg-1 monocrotaline),blank liposomes group(model rats inhaled 150 μL of blank liposomes),sildenafil citrate liposomes group(model group rats inhaled 3 mg·kg-1·d-1 150 μL of sildenafil citrate liposomes),experimental-L group(model group rats inhaled 1 mg·kg-1·d-1 50 μL of aldenafil citrate liposome),experimental-H group(model group rats inhaled 3 mg·kg-1·d-1150 μL of aildenafil citrate liposomes),continuous administration for 3 weeks.At the end of the experiment,the right ventricular systolic pressure of each group of rats was measured by right heart catheterization,and the right heart hypertrophy index,organ index and pulmonary artery wall thickness index were measured.Results The right ventricular systolic pressure(RVSP)of the normal group,the model group,the blank liposomes group,the sildenafil citrate liposomes group,the experimental-L group and the experimental-H group were(26.23±0.60),(53.93±10.20),(50.01±6.93),(32.64±2.62),(30.12±1.98)and(33.45±2.54)mmHg,respectively;RVHI were 0.19±0.03,0.39±0.06,0.37±0.05,0.33±0.03,0.20±0.03 and 0.30±0.04,respectively;the lung indexes were(0.45±0.03)%,(0.85±0.07)%,(0.85±0.15)%,(0.75±0.11)%,(0.47±0.02)%and(0.71±0.09)%,respectively;the WT%were(50.79±10.12)%,(73.25±9.83)%,(72.65±10.09)%,(61.86±2.62)%,(54.68±8.11)%and(56.16±10.83)%,respectively.The model group was compared with the normal group,and the experimental-L group was compared with the model group,the differences were statistically significant(all P<0.05).Conclusion Aildenafil citrate liposomes for inhalation can reduce pulmonary artery pressure,improve right ventricular hypertrophy and pulmonary vascular remodeling in MCT-induced PAH rats,and achieve anti-PAH effect.关键词
