摘要

Abstract

Objective To investigate the mechanism of Chaihu-Shugan-San(CSS)in ameliorating non-alcoholic fatty liver dis-ease(NAFLD)with liver stagnation and Qi stagnation pattern through modulation of gut microbiota and enhancement of intestinal mucosal barrier function.Methods A rat model of NAFLD with liver stagnation and Qi stagnation pattern was established by high-fat diet feeding combined with limb binding.Ninety SPF-grade SD rats were randomly divided into a normal control group(15 rats)and a model preparation group(75 rats).After successful modeling,the model group rats were randomly divided into five subgroups:the NAFLD model group,the Western medicine group(fenofibrate 20mg/kg),the low-dose CSS group(2.5g/kg),the medium-dose CSS group(5g/kg),and the high-dose CSS group(10g/kg).The normal control group received saline administration by gavage daily for 8 weeks.Rats were sacrificed,and serum,liver,and colon tissues were collected for detection of serum biochemical parameters,liver histopathology,colonic immune markers,and gut microbiota composition(analyzed by 16S rRNA gene sequencing).Results CSS significantly reduced serum levels of TG,TC,LDL,ALT,and AST in NAFLD rats(P<0.05).HE staining showed that CSS alleviated hepatic steatosis and reduced pathological injury,with decreased lipid droplets in hepato-cytes.Immunohistochemical analysis indicated that CSS increased the expression of occludin and ZO-1 proteins in the colon tis-sue.16S rRNA gene sequencing revealed that CSS modulated the diversity and composition of gut microbiota,showing similarity to the normal control group.Conclusion Chaihu-Shugan-San significantly ameliorates NAFLD with liver stagnation and Qi stagnation pat-tern by regulating gut microbiota and enhancing intestinal mucosal barrier function.
