可表达禽腺病毒Fiber2基因的重组马立克病病毒疫苗株的构建
Construction of a recombinant Marek's disease virus vaccine strain expressing the Fiber2 gene of fowl adenovirus

Abstract
To develop a recombinant Marek's disease virus(MDV)live vector vaccine for avian hepati-tis-hydropericardium syndrome(HHS),the Fiber2 gene was amplified from a virulent serotype 4 fowl adeno-virus(FAdV-4)strain using PCR and cloned into the pCDNA3.1 vector to construct the recombinant eukary-otic expression plasmid pSV40-Fiber2.Subsequently,the expression fosmid pMS-Fiber2 was constructed through PCR and homologous recombination.This fosmid,along with four other overlapping plasmids covering the entire MDV genome,was co-transfected into chicken embryo fibroblast(CEF)cells to rescue the recombinant virus rMDV-Fiber2.Following serial passages in CEF,rMDV-Fiber2 was identified using PCR,indirect immunofluorescence assay and Western-blot,and its in vitro replication characteristics were analyzed by growth curve plotting.The results indicated that rMDV-Fiber2 stably expressed the Fiber2 protein and exhibited same replication capacity in CEF comparable to the parental virus rMSΔmeq.Challenge experiments demonstrated that chickens immunized with rMDV-Fiber2 achieved an 80%survival rate when challenged with the virulent FAdV-4 strain.In conclusion,the replication ability of the successful constructed rMDV-Fiber2 was similar to that of the parental strain,which provided a foun-dation for developing a recombinant MDV live vector vaccine for HHS.关键词
