Abstract

To develop a universal detection method for goose parvovirus(GPV)and Muscovy duck parvovirus(MDPV),this study designed a set of specific primers targeting the conserved sequence of the NS gene from both GPV and MDPV.A loop-mediated isothermal amplification(LAMP)method and a LAMP-hydroxy naphthol blue(HNB)visual detection method were optimized and established for waterfowl parvovirus detection.The specificity and sensitivity of both methods were evaluated.Additionally,10 clinical waterfowl samples were tested using conventional PCR,LAMP,and LAMP-HNB to assess the practical application of the methods.The results showed that the optimal reaction conditions for the LAMP method were 65℃for 60 minutes.Both methods specifically amplified GPV and MDPV,while no amplification was observed for five other common waterfowl viruses.The minimum detection limit for both methods was 1×10⁻⁵ ng/μL.Moreover,the results of LAMP and LAMP-HNB were 100%consistent with conventional PCR.These findings indicate that the established LAMP and LAMP-HNB visual detection methods exhibit high sensitivity,strong specificity,minimal instrumentation requirements,and easy visualization,making them suitable for rapid on-site detection of waterfowl parvoviruses.
