Effects of Platelet-Rich Plasma on Inflammatory and Catabolic Response Related Factors in Equine Articular Cartilage Explants

Abstract

To explore the efficacy of platelet-rich plasma(PRP)in alleviating inflammation and catabolic responses in cartilage tissue,this study collected and cultured healthy equine articular cartilage explants.Osteoarthritis(OA)-like changes were induced in vitro by the addition of interleukin-1β(IL-1β).The samples were divided into six experimental groups:Negative control,Positive control(triamcinolone acetonide-treated),FP-PRP(fresh pure platelet-rich plasma),DP-PRP(freeze-dried pure platelet-rich plasma),FL-PRP(fresh leukocyte-rich platelet-rich plasma),and DL-PRP(freeze-dried leukocyte-rich platelet-rich plasma).Quantitative real-time polymerase chain reaction(qPCR)and enzyme-linked immunosorbent assay(ELISA)were used to assess inflammation and metabolic markers in both the culture medium and cartilage cells.The results showed that compared with the Negative control group,the FP-PRP,DP-PRP,and FL-PRP groups had reduced mRNA expression of cyclooxygenase-2(COX-2),prostaglandin E synthase(PTGES),and cartilage oligomeric matrix protein(COMP),though the differences were not statistically significant(P>0.05).However,the expression of collagen type II α1 chain(COL2A1)was significantly decreased(P<0.05),and glycosaminoglycan(GAG)levels in the medium were significantly reduced(P<0.01 or P<0.001).Matrix metalloproteinase-13(MMP-13)expression showed an upward trend in the PRP-treated groups without significance(P>0.05),except in the DL-PRP group where MMP-13 expression was significantly increased(P<0.05).These findings suggest that FP-PRP and DP-PRP can attenuate inflammation in the equine OA model,whereas DL-PRP shows limited anti-inflammatory effect.PRP can positively regulate the metabolic balance of the OA model,showing potential to mitigate cartilage degeneration in equine cartilage explants.
