Infection and Species Identification of Cryptosporidium spp.in Domestic Peacocks(Pavo cristatus)in Zhongmu,Henan Province

Abstract
Cryptosporidium infection is relatively common in peacock farming.To investigate the infection status and genetic characteristics of Cryptosporidium in peacocks in Zhongmu County,Henan Province,a total of 190 fecal samples were collected from three peacock farms.Morphological examination and PCR amplification targeting the 18S rRNA gene were performed.Positive samples were sequenced,and a phylogenetic tree was constructed to determine the evolutionary relationship between the isolates and known Cryptosporidium species.The results showed that 9 out of 190 fecal samples were positive for Cryptosporidium,with a positivity rate of 4.74%.Based on flotation using saturated sugar solution,the oocysts were preliminarily identified as Cryptosporidium galli.Infections were detected in 2 of the 3 surveyed farms,with infection rates of 7.58%and 6.35%,respectively;no significant difference was found among farms(P>0.05).Cryptosporidium infection was observed only in peacocks under 90 days of age,with a positivity rate of 9.78%(9/92),while no infections were found in peacocks aged 91-360 days or over 361 days.Infection rates differed significantly among age groups(P<0.01).No correlation was observed between Cryptosporidium infection and diarrhea symptoms(P>0.05).The 9 positive isolates were grouped into two sequence types(HK1 and HK2),showing 96.1%and 100%similarity to C.galli,respectively.Phylogenetic analysis confirmed both sequences clustered within the C.galli clade,showing the highest homology with avian-origin C.galli isolates in China.This is the first report of C.galli infection in peacocks in China,expanding the known diversity of Cryptosporidium species in birds and providing a theoretical basis for further studies on the transmission characteristics of C.galli.关键词
