Effect of Xuezhikang on Hyperlipidemia in Golden Hamsters by Regulating Gut Microbiota

Abstract
Objective To investigate the effects of Xuezhikang on lipid levels in a hyperlipidemia model of golden hamsters and explore the potential mechanisms involved.Methods Forty adult male golden hamsters were randomly divided into five groups(with 8 mice in each group):normal control(equal volume of physiological saline),model(equal volume of physiological saline),simvastatin(100 mg/kg),low-dose Xuezhikang(25 mg/kg),and high-dose Xuezhikang(100 mg/kg).All groups were fed the high-fat diet for four weeks to induce hyperlipidemia.Drugs or physiiological saline were administered by gavage once daily for four weeks.Serum levels of total cholesterol(TC),triglycerides(TG),low-density lipoprotein cholesterol(LDL-C),and high-density lipoprotein cholesterol(HDL-C)were measured by the automatic biochemical analyzer.Reactive oxygen species(ROS)were measured by fluorescence quantification,and the levels of malondialdehyde(MDA),superoxide dismutase(SOD),and glutathione peroxidase(GSH-Px)were also determined.The levels of interleukin interleukin-1β(IL-1β),interleukin-6(IL-6),and tumor necrosis factor-α(TNF-α)were detected by the enzymelinked immunosorbent assay.Colonic contents were subjected to high-throughput sequencing to analyze fecal microbiota.Operational taxonomic unit were clustered at 97%similarity using USEARCH(version 7.1),and chimeras were removed.Principal coordinate analysis and partial least squares discriminant analysis were used to evaluate the overall structure and community features of the gut microbiota.Results Compared with those in the model group,the levels of TC,ROS,MDA,IL-1β,IL-6,and TNF-α significantly decreased and the levels of HDL-C significanely increased in the simvastatin and both Xuezhikang groups.TG levels significantly reduced in the high-dose Xuezhikang group,LDL-C level significantly decreased,SOD and GSH levels increased both in the simvastatin and high-dose Xuezhikang groups(P<0.05 or P<0.01).Compared with those in the model group,community richness and gut microbial diversity significantly increased in the high-dose Xuezhikang group(P<0.01).At the terms of gut microbiota composition,the proportion of Lachnospiraceae_NK4A136_group was significantly increased,while the proportion of Lactobacillus was significantly reduced in the high-dose Xuezhikang inccveased group(P<0.01).Conclusion Xuezhikang can alleviate lipid metabolism disorders in golden hamsters with hyperlipidemia,possibly by regulating gut microbiota composition and improving oxidative stress and inflammatory responses.关键词
