芝麻酚对对乙酰氨基酚致肝损伤模型小鼠的保护作用及其机制
Protective Effect and Mechanism of Sesamol on Acetaminophen-Induced Liver Injury Model Mice

Abstract
Objective To investigate the effect of sesamol on liver injury in model mice.Methods A total of 60 KM mice were randomly divided into control group[equal volume of 0.5%carboxymethyl cellulose sodium(CMC-Na)solution],model group(equal volume of 0.5%CMC-Na solution),glutathione group(200 mg/kg),and high-dose,medium-dose and low-dose sesamol groups(200,100,50 mg/kg),with 10 mice in each group.The corresponding drugs or CMC-Na were administered by gavage once a day for 7 days.After the last administration,the acetaminophen(300 mg/kg)was intraperitoneally injected to replicate a liver injury mouse model.The pathological morphology of liver tissue was observed by hematoxylin-eosin staining.The levels of alanine aminotransferase(ALT),aspartate aminotransferase(AST),alkaline phosphatase(AKP),total bilirubin(TBiL),superoxide dismutase(SOD),catalase(CAT),and malondialdehyde(MDA)in plasma were determined.Protein expression levels of nuclear factor erythroid 2-related factor 2(Nrf2),phosphorylated and non-phosphorylated nuclear factor-κB p65(p-NF-κB p65/NF-κB p65),tumor necrosis factor-α(TNF-α),and interleukin-6(IL-6)in the liver omogenate were determined by Western blotting.Results In the control group,hepatocytes and nuclear structures exhibited normal morphology with clear boundaries.In the model group,hepatocytes were swollen,and partial nuclear dissolution was observed.The degree of hepatocyte swelling and inflammatory cell infiltration was alleviated in the high-dose,medium-dose and low-dose sesamol group.Compared with those in the model group,the levels of ALT,TBiL and MDA in plasma,as well as the protein expression levels of IL-6 and TNF-α in liver homogenate were significantly decreased in the high-dose,medium-dose and low-dose sesamol groups(P<0.05 or P<0.01),while the protein expression level of Nrf2 was significantly increased(P<0.05 or P<0.01).The plasma levels of SOD and AST in the high-dose and medium-dose sesamol groups were significantly increased(P<0.05 or P<0.01)and the p-NF-κB p65/NF-κB p65 protein in the high-dose and medium-dose sesamol groups were significantly decreased(P<0.05 or P<0.01).The plasma CAT level in the high-dose sesamol group was significantly increased(P<0.01).Conclusion Sesamol can regulate Nrf2/NF-κB signal pathway for acetaminophen-induced liver injury in mice,to enhance antioxidant and anti-inflammatory activities.关键词
