电针干预改善大脑中动脉栓塞大鼠脑部缺血机制的研究
Electroacupuncture intervention on mechanism of cerebral ischemia in rats with middle cerebral artery occlusion(MCAO)

Abstract
Objective:To observe the effect of electroacupuncture on the expression of HtrA1 in astrocytes within the hippocampal tissue of rats with acute cerebral infarction.Methods:Twenty-four male SD rats were selected and randomly divided into the sham-operation,model,and electroacupuncture groups,with 8 rats in each group.The model and electroacupuncture groups underwent right middle cerebral artery occlusion(MCAO)modeling via the intraluminal filament occlusion method.The sham-operation group received identical surgical exposure without filament insertion.Twenty-four hours after successful modeling,rats in the electroacupuncture group received electroacupuncture treatment at Baihui(GV20)and Shenting(GV24)using dense-disperse waves with alternating frequencies of 2/15 Hz and a current intensity of 1.5-2 mA.Cerebral blood flow changes,astrocyte expression and activation status,HtrA1 mRNA levels,HtrA1 protein expression levels,and TNF-α,IL-1β,and IL-6 levels of three groups were measured and compared seven days after the operation.Results:Western blotting showed that HtrA1 expression in the model group was significantly increased compared to the sham-operation and electroacupuncture groups(both P<0.05).Quantitative real-time PCR(qRT-PCR)showed that HtrA1 mRNA expression level in the electroacupuncture group was significantly lower than that in the model group(P<0.05).ELISA showed that the model group had the elevated levels of TNF-α,IL-1β,and IL-6 compared to the sham-operation group(all P<0.05),while the electroacupuncture group had the reduced levels of TNF-α,IL-1β,and IL-6 compared to the model group(all P<0.05).Conclusions:Electroacupuncture can alleviate injury,improve neurological function,enhance cerebral blood flow,and suppress the inflammatory response,and the mechanism may be associated with the inhibition of HtrA1 protein expression and suppression of astrocyte polarization.关键词
