T2mapping对止痛健骨方防治兔膝骨关节炎关节软骨衰老疗效的监测
Effect monitoring of Zhitong Jiangu Formula in prevention and treatment of articular cartilage senescence in rabbit knee osteoarthritis by T2 mapping

Abstract
Objective:To evaluate the therapeutic effect of Zhitong Jiangu Formula(ZTJGF)in preventing and treating the senescence of articular cartilage in knee osteoarthritis(KOA),and to explore the value of T2 mapping in monitoring the therapeutic effect.Methods:A KOA model in New Zealand white rabbits was produced by immobilizing the knee joint in plaster bandage in full extension.According to different treatment methods,32 New Zealand white rabbits were randomly divided into the normal control group,the model control group,the ZTJGF treatment group,and the glucosamine sulfate treatment group,with 8 rabbits in each group.After two weeks of immobilizing,the normal control group and the model control group were given the same volume of distilled water by gavage,while the ZTJGF treatment group was given ZTJGF,and the glucosamine sulfate treatment group was given sulfuric acid and glucose capsules.After 4 weeks of treatment,knee T2 mapping was performed on all groups,and the expression of p16INK4a in articular chondrocytes was analyzed by immunohistochemistry.The differences in T2 values of articular cartilage and p16INK4a expression among the four groups were compared.The correlations among different intervention methods,T2 values and p16INK4a expression were analyzed.Results:T2 value in the ZTJGF treatment group and the glucosamine sulfate group were higher than that in the normal control group,but lower than that in the model control group(all P<0.05).The p16INK4a expression in the ZTJGF treatment group,the glucosamine sulfate group and the normal control group had no differences(P>0.05),but were significantly lower than that in the model control group(all P<0.05).p16INK4a expression was nonlinearly related to T2 value and was slowly increased and then rapidly increased as the T2 value increased(P<0.05).Conclusions:The ZTJGF can prevent and treat the aging of articular cartilage in KOA,and T2 mapping can be used as a non-invasive assessment tool for prevention of cartilage senescence.关键词
