蒙药当贡-3抗心肌缺血再灌注损伤分子机制的网络药理学研究
Network Pharmacology Study on the Molecular Mechanism of Mongolian Medicine Danggong-3 in Anti-Myocardial Ischemia-Reperfusion Injury

Abstract
Objective:To explore the material basis and molecular mechanism of myocardial ischemia-reperfu-sion injury(MI/RI)based on network pharmacology and molecular docking.Methods:The chemical components of Mongolian medicine Danggong-3(DG3)were collected through literature research and the TCMSP database.The PharmMapper database was used to predict DG3 component targets.MI/RI transcriptome data(GEO ID:GSE103731)were obtained from the GEO database,and differentially expressed genes were calculated using the Limma package.Venn analysis was employed to screen potential therapeutic targets of DG3 in protecting against MI/RI.Protein-Protein Interaction(PPI)analysis was conducted to identify core targets of DG3 in MI/RI protec-tion.Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)enrichment analyses were per-formed to determine key pathways associated with the potential targets.Results:A total of 29 DG3 chemical com-ponents and 731 drug targets were identified.Differential analysis revealed 2 202 MI/RI-related targets,and Venn analysis identified 247 potential therapeutic targets of DG3 for MI/RI protection."Herb-Component-Common Target"network analysis identified key components such as Ellagic acid,Liquiritigenin,Hyoscyamine,and Auran-tio-obtusin,etc.PPI analysis revealed TNF,Akt1,TP53,and SRC as potential core targets of DG3 in MI/RI protec-tion.GO enrichment analysis mainly involved responses to hormones,responses to peptides,positive regulation of phosphorus metabolic processes,and protein kinase activity.KEGG enrichment analysis showed that these poten-tial targets were strongly associated with the PI3K/Akt signaling pathway.Conclusion:DG3 may exert protective effects against MI/RI via proteins such as TNF,Akt1,TP53,and SRC,possibly through the PI3K/Akt signaling pathway.关键词
