Abstract

Objective:To investigate the effect of Tianxiangdan on the protein expression levels of apoptosis related factors heat shock protein 27(HSP27)and p38 mitogen-activated protein kinase(p38MAPK)in myocardi-al ischemia model rats,and to reveal the molecular mechanism of Tianxiangdan in regulating apoptosis and treat-ing myocardial ischemia injury.Methods:A rat model of myocardial ischemia was established by ligation of the left anterior descending coronary artery and randomly divided into 6 groups,with 12 rats in each group.After suc-cessful model establishment,low,medium,and high doses of Tianxiangdan granules(1.1,3.4,and 10.2 g/kg,re-spectively)were administered.Changes in cardiac function were evaluated using electrocardiography(ECG),histo-pathological changes in myocardial tissue were observed using HE staining,and the expression levels of HSP27 and p38MAPK were detected by western blotting.Results:ECG results showed significant elevation of ST seg-ments and T waves in the model group after surgery compared with before surgery levels(P<0.05),indicating suc-cessful establishment of the myocardial ischemia injury model.Compared with the model group,all dose groups of Tianxiangdan significantly reduced ST segment elevation after 7 days of intervention(P<0.05).HE staining showed that myocardial fibers in the blank and sham operation groups were neatly arranged without obvious patho-logical changes.In contrast,the model group exhibited significant myocardial fiber rupture,dissolution necrosis,vacuolar degeneration,and interstitial fibrous tissue proliferation.These pathological changes were alleviated to varying degrees in all Tianxiangdan.Western blotting analysis revealed that the expression level of HSP27 protein was significantly higher in the model group and all Tianxiangdan groups compared with the blank and sham opera-tion groups(P<0.05).Moreover,the high-dose Tianxiangdan group showed significantly higher HSP27 expres-sion than the low-dose group(P<0.05).No significant differences were observed in the expression levels of p38MAPK among the groups(P>0.05).Conclusion:Tianxiangdan significantly increases the expression level of HSP27 protein in myocardial tissue of rats with myocardial ischemia injury,which may be a key molecular target for Tianxiangdan to regulate apoptosis and improve myocardial ischemia injury.
