通督灸调节Keap1/Nrf2/ARE信号通路对急性脑缺血再灌注大鼠氧化应激损伤的影响
Effect of Tongdu Moxibustion on Oxidative Stress Injury in Acute Cerebral Ischemia-Reperfusion Rats by Regulating the Keap1/Nrf2/ARE Signaling Pathway

Abstract
Objective:To investigate the effect of Tongdu moxibustion on oxidative stress injury in acute cere-bral ischemia-reperfusion(CIR)rats by regulating the Keap1/Nrf2/ARE signaling pathway.Methods:90 SD rats were divided into sham operation group,model group,model+Nrf2 inhibitor all-trans retinoic acid(ATRA)group(10 mg/kg),edaravone group(3 mg/kg),Tongdu moxibustion group,and Tongdu moxibustion+ATRA group(10 mg/kg).Neurological function was assessed using Longa scoring criteria,and brain infarct volume was measured by TTC staining.Pathological changes in the ischemic cortex were observed by HE staining.Superoxide dismutase(SOD)activity and malondialdehyde(MDA)content in serum were measured using the WST-8 colorimetric meth-od and thiobarbituric acid(TBA)method,respectively.Reactive oxygen species(ROS)levels in brain tissue were analyzed using the 2',7'-dichlorofluorescin diacetate(DCFH-DA)staining method.Protein expression levels of Keap1,Nrf2,HO-1,and NQO1 in the ischemic cortex were detected by Western blotting.Results:Compared with the model group,the Tongdu moxibustion group showed significantly reduced pathological changes in the ischemic cortex,decreased neurological deficit scores,infarct volume percentage,MDA content,and Keap 1 protein expres-sion,while increased SOD activity,nuclear Nrf2/Nrf2,HO-1,and NQO1 protein expression(0.05).After ATRA in-tervention,the therapeutic effects of Tongdu moxibustion on acute cerebral ischemia-reperfusion rats were weak-ened(P<0.05).Conclusion:Tongdu moxibustion has a significant effect on reducing oxidative stress injury in acute cerebral ischemia-reperfusion rats,which may be related to the upregulation of the Keap1/Nrf2/ARE signal-ing pathway.关键词
