补阳还五汤对脑缺血再灌注损伤模型大鼠肠道菌群的影响
Effect of Buyang Huanwu Decoction on Intestinal Flora in Rats with Cerebral Ischemia-Reperfusion Injury Model

Abstract
Objective:To investigate the effect of Buyang Huanwu Decoction on the intestinal flora in a rat mod-el of cerebral ischemia-reperfusion injury.Methods:Genomic DNA was extracted from fecal samples using the OMEGA Soil DNA Kit.The V3～V4 hypervariable regions(about 468 bp)of bacterial 16S rRNA genes were ampli-fied by PCR using specific primers 338F(5'-barcode-ACTCCTACGGGAGGCAGCA-3')and 806R(5'-GGAC-TACHVGGGTWTCTAAT-3').Libraries were constructed using the TruSeq Nano DNA LT Library Prep Kit,fol-lowed by quality control and paired-end sequencing(2×250 bp)for qualified libraries.Results:1)The model group showed obvious cerebral infarction,which was significantly reduced after administration of the decoction.2)Significant changes were observed in the structure of the gut microbiota among groups.At the genus level,Lactoba-cillus was most abundant in the control group,while its abundance was significantly decreased in the model group,accompanied by an increase in Akkermansia.In the treatment group,Bacteroides and Allobaculum remained domi-nant,but the abundances of Lactobacillus and Bifidobacterium were notably increased.3)Although some ASVs/OTUs overlapped among different groups,each group also had unique sequences.For example,the control group had 4 648 unique ASVs/OTUs,the model group had 2 126,and the treatment group had 2 308.In the treatment group,Clostridium was the main contributor to the significant differences compared with the other two groups.In the model group,Enterococcaceae contributed most to the differences,while in the control group,Firmicutes was the key differential factor.4)The control group partially overlapped with the model group in a few functional units,but only a small number of functional units overlapped among all three groups.Moreover,both the model and treat-ment groups were far from the control group in clustering analysis.Conclusion:Buyang Huanwu Decoction pro-motes the regeneration of intestinal Lactobacillus and Bifidobacterium,enhancing their probiotic functions and mak-ing them more concentrated.关键词
