低温处理牡丹试管苗前后的转录组分析
Transcriptome Analysis of Tree Paeonia suffruticosa Plantlet before and after Low Temperature Treatment

Abstract
The main reason why the tissue culture technology of Paeonia suffruticosa has not yet been applied to industrialization is that the above-ground parts of plantlet enter dormancy after rooting,resulting in a low survival rate after transplantation.Research has found that treating tree peony pla-ntlets at a low temperature of 4℃ for 60 days could promote the germination of the apical bud and break the dormancy of the apical bud.In order to analyze the related mechanism of low-temperature treatment on the apical bud germination of plantlet,phenotypic observation and transcriptome sequencing were conducted on plantlet treated with low temperature for different days.The results indicated that after the rooting plantlet were treated with low temperature,the ABI transcription was inhibited,and the expressions of some key genes such as PP2C,PYL,and ARF in the ABA synthe-sis pathway were suppressed,hindering ABA biosynthesis.At the same time,the expressions of some key genes GA2ox1 and GAI in the GA biological pathway were activated,promoting GA syn-thesis led to the balance of GA and ABA contents in plantlet seedlings,promoting the germination of apical buds.关键词
