Abstract

In order to study the physiological characteristics of Bupleurum scorzonerifolium seeds,the seed quality,storage substance content and antioxidant enzyme activity of six groups of Bupleurum scorzonerifolium seed samples from two production areas were determined.The changes in storage substance content and antioxidant enzyme activity during different pretreatment methods and the seed germination process were compared through tissue culture experiments.The results showed that the quality of Bupleurum scorzonerifolium seeds had a significant impact on the content of stored sub-stances and the activity of antioxidant enzymes.Ultrasonic treatment for 30 minutes could significant-ly increase the germination rate of Bupleurum scorzonerifolium seeds.Soaking seeds in gibberellin at a concentration of 100 mg/L was optimal,which could break the dormancy of Bupleurum scorzoneri-folium seeds and increase the germination rate to 41.33%,while significantly enhancing the POD ac-tivity.Different disinfection methods had a significant effect on the oxidase activity of Bupleurum scorzonerifolium seeds.The germination process showed an insignificant stage.The contents of total protein and soluble sugar were the highest on the first day and then gradually decrease to a stable lev-el.The CAT activity decreased with the increase of dark culture time,while the SOD activity first in-creases and then decreased.The first peak period of germination corresponds to the time point when the stored substances and enzyme activities in the seeds decline,indicating that the germination envi-ronment at this stage and before was crucial for seed germination.The seeds of Bupleurum scorzoner-ifolium had obvious physiological dormancy characteristics.Exogenous gibberellin could effectively in-crease the germination rate by breaking dormancy and regulating enzyme activity.
