悬铃木LFY基因第二内含子多态性分析
Polymorphism Analysis of the Second Intron of LFY Gene in Platanus acerifolia

Abstract
The Platanus acerifolia is a hybrid of the Platanus occidentalis L.and the Platanus orientalis L.,showing obvious hybrid vigor and is widely used in landscaping.The LFY gene is a rare single-copy gene in dip-loid angiosperms.To explore the polymorphism of the second intron of the LFY gene in Platanus acerifolia,eight Platanus acerifolia samples were used as materials.Two pairs of primers,S257/S258 and S262/S263,were designed to amplify partial fragments of the second intron of the LFY gene respectively,and nucleotide sequence polymorphism analysis was conducted.The results indicated that based on SNP loci,primers S257/S258 could distinguish between Yuanke115,Yuanke23 and Sanqiu,and primers S262/S263 could distinguish between Yuanke23 and Sanqiu.In addition,a pair of SSR primers was developed for the 3′end GA repeat site of the second intron of the LFY gene in the Platanus occidentalis L.and was detected by capillary electrophoresis.The two LFY sequences of Yuanke10,Yuanke22 and Fast-growing Plane trees were respectively from the Pla-tanus occidentalis L.and Platanus orientalis L..The LFY of Yuanke115 and Yuanke23 both came from the Pla-tanus occidentalis L.,and the LFY of Yuanke12 all came from the Platanus orientalis L..关键词
