Value of injectable thermosensitive hydrogel encapsulating chalcone derivatives-compound 5 for local chemotherapy of breast cancer

Abstract
Objective To explore the value of chitosan(CS)/β-glycerophosphate(β-GP)hydrogel system loaded with chalcone derivative-compound 5(chalcone derivatives-compound 5,CHAD)for local treatment of breast cancer,and to evaluate the antitumor activity and biosafety of this system.Methods CHAD was incorporated into CS/β-GP,denoted as CS/β-GP@CHAD.This hydrogel system was measured in terms of its temperature sensitivity,microstructure,rheological properties,and drug release performance.Subsequently,cell counting kit-8,flow cytometry,and hematoxylin and eosin staining were utilized to assess its biosafety.Finally,a model of xenograft tumor in mice was established to evaluate the antitumor efficacy of CS/β-GP@CHAD in vivo.Results The CS/β-GP@CHAD hydrogel underwent a remarkable transformation:from a colorless,transparent liquid at 20℃ to an irregular,porous gel at 37℃,with a phase transition time of 65 s and a phase transition temperature of 36 ℃.This system excels in delivering drugs with sustained-release properties at body temperature.CS/β-GP@CHAD significantly promoted apoptosis of MCF-7 cells(human breast cancer cells),while had minimal impact on the physiological activity of MCF-10A cells(human normal breast epithelium cells),and it exhibited no toxicity to mouse organs.Tumor transplantation experiment triumphantly demonstrated that the anti-breast cancer efficacy of CS/β-GP@CHAD surpassed that of free CHAD when administered at equivalent doses.Conclusion The encapsulation of CS/β-GP can effectively enhance the bioavailability of CHAD,potentiate its anti-breast cancer activity,and demonstrate favorable biosafety,potentially emerging as a promising drug delivery system for breast cancer treatment.关键词
