摘要

Abstract

The purpose of this study was to prepare polysaccharide liposomes from Schisandra chinensis(SPL)by reversed-phase evaporation,optimize the preparation process,and research its characterization and in vitro drug release performance.The single factor test was carried out on four factors that affected the encapsulation efficiency of SPL,including the ratio of membrane to material,the ratio of lipid to drug,ultrasonic time and water bath temperature.Based on the premise of single factor test results,the Design Expert 10 software was used to establish a mathematical model to predict the optimal combination of process parameters,and the feasibility was verified by the experiment.The physical stability of SPL was evaluated by visual observation,the structural integrity of vesicles was analyzed by transmission electron microscopy,the particle size distribution and Zeta potential were determined by nanoparticle tracking analyzer,the polydispersity index(PDI)was detected by nanoparticle size analyzer,and the chemical structure integrity was detected by Fourier Transform Infrared Spectroscopy(FTIR).In addition,the transmembrane permeability properties of liposomes were assessed by in vitro drug release assays.The results showed that the optimal process parameters for the preparation of SPL by reversed-phase evaporation were the membrane-to-material ratio of 2.29∶1(m∶m),the lipid-to-drug ratio of 15.78∶1(m∶m),the ultrasonic time of 9 min,and the water bath temperature of 45℃.Under these conditions,the predicted encapsulation rate of SPL was 74.00%,the verification value was 72.90%,and the relative deviation between the theoretical value and the actual value was 1.51%.Through the characterization of SPL,it was found that SPL was a translucent suspension in the visual state,with uniform texture and no precipitation.TEM observation showed that SPL had complete morphology,clear contour and uniform size.The average particle size of the liposomes was 154.9 nm,the Zeta potential was(-46.76±0.60)mV,and the PDI was 0.22.FTIR analysis confirmed the integrity of the phospholipid bilayer and the absence of chemical bond cleavage between polysaccharides and liposomes.In vitro release assay showed that liposomes could effectively delay the release rate of drugs as carriers compared with Schizandra chinensis polysaccharides.In this study,the optimal preparation process for polysaccharide liposomes from Schisandra chinensis was obtained and the liposomes were well characterized,the in vitro drug release properties showed that they had the potential to improve bioavailability,which could provide a reference for subsequent functionalization studies.
