Chinese herbal feed additives for the prevention of diarrhea in lambs:a clinical trial

摘要

Abstract

This trial aimed to investigate the preventive effect of a Chinese herbal feed additive on diarrhea in suckling lambs.Fifty-six healthy 7-day-old Hu lambs(approximately 5 kg body weight)were selected and randomly allocated into four groups:a control group,and trial groupsⅠ,Ⅱ,and Ⅲ.The groups were supplemented with 0%,1%,2%,and 4%of a compound Chinese herbal additive(composed of Zingiber officinale,Pueraria montana var.lobata and Aucklandia costus,etc)in their starter feed,respectively,for a 45-day feeding period.Diarrhea incidence was observed and recorded daily to calculate the diarrhea rate.Body weight and feed intake were measured every 15 days to calculate average daily gain(ADG)and feed conversion ratio(FCR).Blood samples were collected concurrently every 15 days;plasma was separated and used to measure biochemical indices(TP,ALB,ALT,CRE)on a microplate reader using enzymatic methods(bromocresol green method,BCA method,colorimetric method and sarcosine oxidase method).Antioxidant parameters(GSH-Px,T-AOC,SOD,MDA)were determined on a microplate reader/spectrophotometer using specific assay principles(visible chromogenic method,FRAP method,WST-8 method and microplate/TBA method).Immune markers(IgG,IFN-γ,IL-1β,IL-6,IL-10,TNF-α)were quantified using a double-antibody sandwich enzyme-linked immunosorbent assay(ELISA)with specific commercial kits and a microplate reader.The results showed that the diarrhea rates of lambs in group Ⅱ and Ⅲ were extremely significantly lower than that in the control group(P<0.01);there were no significant differences in body mass,feed intake,average daily gain,or feed conversion ratio among the groups(P>0.05),but groupⅡshowed relatively better performance in these indicators.On day 45,plasma ALB levels in all three experimental groups were extremely significantly higher than that in the control group(P<0.01),and ALT levels were significantly lower than that in the control group(P<0.05).On day 15,significant changes in plasma antioxidant indices began to appear in the experimental groups compared to the control group.On day 45,GSH-Px,SOD,T-AOC and MDA in all three experimental groups differed significantly from the control group(P<0.05),with groupⅡ being significantly better than groups Ⅰ andⅢ.On day 15,30 and 45,plasma IgG and IFN-γ levels in all three experimental groups were extremely significantly higher than that of the control group(P<0.01),the anti-inflammatory factor IL-10 level in group Ⅱ was extremely significantly higher than that of the other groups(P<0.01),while the levels of pro-inflammatory factors(IL-1β,IL-6,TNF-α)were extremely significantly lower than that of the other groups(P<0.01).Therefore,adding this Chinese herbal feed additive to lamb starter feed can enhance antioxidant function and regulate immune function,thereby reducing the diarrhea rate without affecting diet palatability or feed intake.A 2%inclusion level is recommended as the optimal dietary dose.These results provide a scientific basis for the development of Chinese herbal additives to prevent lamb diarrhea.
