小反刍兽疫诊断技术研究进展
Review on diagnosis technology of peste des petits ruminants

Abstract
Peste des petits ruminants(PPR)is a highly contagious infectious disease caused by peste des petits ruminants virus(PPRV),which is characterized by fever,stomatitis and enteritis,and seriously endangers domestic and wild small ruminants.In recent years,the PPR epidemic in China is generally stable,but the risk of being transmitted from neighboring countries persists.Rapid and accurate diagnostic techniques are essential for timely detection and blocking the spread of the epidemic.Currently widely used diagnostic methods for PPR virus can be categorized into etiological detection and serological detection.Etiological detection mainly includes virus isolation,reverse transcription polymerase chain reaction(RT-PCR),loop-mediated isothermal amplification(RT-LAMP),recombinase polymerase amplification(RT-RPA)and recombinase aided amplification(RT-RAA).Among them,virus isolation and culture is the gold standard for detecting PPRV,qRT-PCR is considered the gold standard for PPRV nucleic acid detection,and RT-LAMP,RT-RPA and RT-RAA isothermal amplification techniques are suitable for rapid diagnosis in the field.Serological detection includes virus neutralization test(VNT),enzyme-linked immunosorbent assay(ELISA),immunochromatographic strip,chemiluminescence immunoassay methods and time-resolved fluorescence immunoassay techniques.Among them,VNT is the gold standard for antibody detection recognized by the World Organization for Animal Health;the immunochromatographic strip is simple to operate and the results appear quickly,which is suitable for the rapid detection of PPRV.In addition,RT-PCR,qRT-PCR and competitive ELISA are the main detection methods recommended by the Diagnostic Techniques for Peste des Petits Ruminants(GB/T 27982-2011).Different detection techniques have their own advantages and disadvantages,and the inspector can choose the appropriate diagnostic technique according to the specific needs.According to the clinical needs of grassroots personnel and the demands of epidemic surveillance,the future PPR diagnostic technology may develop towards the goals of simple operation,low cost,visualization of results and more efficient,high sensitivity,high specificity and high throughput.关键词
