Transcriptional regulation and detoxification role of mGSTl in spotted-wing drosophila Drosophila suzukii under cyantraniliprole and emamectin benzoate stress

Abstract
To elucidate the role of microsomal glutathione S-transferase(mGST)in the detoxification metabolism of spotted-wing drosophila Drosophila suzukii and its transcriptional regulatory mecha-nism,transcriptome data of D.suzukii were analyzed to identify mGST genes with upregulated expres-sion.Real-time quantitative polymerase chain reaction(qPCR)was utilized to detect its tissue-specific expression patterns.RNA interference(RNAi)experiments were conducted to investigate its role in pes-ticide tolerance.The 5'flanking sequences of the identified genes were cloned and analyzed,and yeast one-hybrid assays were performed to verify transcription factors interacting with this regulatory region.The results demonstrated that mGSTl expression was significantly upregulated following induction by cyantraniliprole,with high expression level in the Malpighian tubules.Under emamectin benzoate treat-ment,mGSTl was highly expressed in the fat body.RNAi-mediated suppression of mGSTl significantly increased the sensitivity of D.suzukii to both insecticides.Moreover,yeast one-hybrid assays confirmed that the transcription factor CROC(crocodile)regulates the expression of mGSTl.These results suggest that mGSTl is involved in the detoxification of cyantraniliprole and emamectin benzoate in D.suzukii,and that its expression is regulated by the transcription factor CROC.关键词
