基于热蛋白质组学技术的三七水煎液治疗脑病作用靶标及潜在新功效研究
Thermal Proteome Profiling Reveals the Targets and Potential New Efficacy of Notoginseng Radix et Rhizoma Decoction in Treating Encephalopathy
摘要
Abstract
Objective:Based on the research model for integrative effect of TCM,which consist of modules including"detecting targets of the drug","targeted-regulated molecule association","obtaining the efficacy with the regulated molecule","clinical validation of efficacy",and"inheritance and innovation of TCM",this study gave precise explanation for the function of the Notoginseng Radix et Rhizoma decoction in brain diseases treatment,,and reveals its potential new efficacy..Methods:Thermal proteome profiling was employed for high-throughput screening of the overall targets of this decoction in the mouse brain tissue.The Ingenuity Pathway Analysis(IPA)database was used for the association analysis of the targets of Notoginseng Radix et Rhizoma decoction and the downstream target proteins.Genvestigator was further used to mine and analyze the target-associated diseases.Results:In this study,15 direct targets and 17 indirect targets of Notoginseng Radix et Rhizoma decoction were screened out.The analyses via IPA and Genvestigator showed that some of these targets were involved in the treatment of ischemic stroke and Alzheimer's disease and they were capable of repairing the nerve damage through the new efficacy of neural regeneration.Conclusion:In this study,we elucidate the mechanism of Notoginseng Radix et Rhizoma decoction in treating encephalopathies from the perspective of target and revealed the potential efficacy of this decoction in repairing nerve damage through nerve regeneration.关键词
