The effect and mechanism of 3-benzyl-5-O-glucoside cycloicariin in improving vascular restenosis after balloon injury in rats

Abstract
Objective To investigate the effect and underlying mechanism of 3-benzyl-5-O-glucoside cycloicariin(designated as:lq210617)on vascular restenosis following balloon injury in rats.Methods The experimental ca-rotid artery balloon injury model was established in rats,which were randomly assigned to five groups:sham oper-ation group,model group and lq210617 treatment groups at doses of 10,20,40 mg/kg.HE staining was used to evaluate the morphological changes of common carotid artery.The expression of vascular smooth muscle cells contractile protein α-SMA,synthetic protein OPN,TLR4,IRAK-1,ERK1/2,p-ERK1/2 and PKC-s were detected by immunohistochemistry.Results The NIA,NIA/MA,and NIA/EELA values in 20 and 40 mg/kg lq210617 groups were significantly lower than those in the model group.α-SMA protein expression was significantly up-regulated and OPN protein expression was significantly down-regulated.the protein levels of TLR4,IRAK-1,ERK1/2,p-ERK1/2,and PKC-s was significantly down-regulated(P<0.05).Conclusion lq210617 effectively suppressed the phenotypic switching of VSMCs in the balloon-injured region and improved vascular restenosis.The underly-ing mechanism involves the suppression of the TLR4/IRAK-1/ERK1/2signaling cascade activation.关键词
