基于网络药理学研究姜黄素抑制SPP1表达诱导舌鳞癌细胞凋亡的机制
The mechanism of curcumin inhibiting SPP1 expression leading to apoptosis in tongue squamous cells based on network pharmacology
摘要
Abstract
Objective To explore whether curcumin intervention is related to the expression of secreted phospho-rylated protein 1(SPP1)in tongue squamous cell carcinoma(TSCC).Methods In this study,differential expres-sion genes(DEGs)was identified through network pharmacological analysis,protein-protein interaction(PPI)network was constructed,and the key genes of TSCC during curcumin treatment was obtained.SPP1 expression level was analyzed according to GREPIA database,and the relationship between the key gene expression and sur-vival was detected by survival analysis.CCK-8 assay,clone formation assay,flow cytometry and western blot were used to verify its effects on cell proliferation,apoptosis and SPP1 expression in Cal27 cells.Results SPP1 was highly expressed and secreted in TSCC tissues,which was positively correlated with lower survival rate in patients with TSCC(P<0.05).SPP1 was the core target of curcumin therapy for TSCC,which was associated with apop-tosis.Meanwhile,curcumin reduced the expression level of SPP1,inhibited its proliferation ability,and induced apoptosis in Cal27 cells(P<0.05).Conclusion Curcumin promoted apoptosis and decreased the expression lev-el of SPP1 in Cal27 cells.关键词
