Mechanism of Icariin in Regulating Apoptosis of Oxygen-Glucose Deprived Neurons

Abstract
Objective:To investigate the possible mechanism of icariin(ICA)in regulating the apoptosis of oxygen-glucose deprived(OGD)neu-rons.Methods:N2a cells(neuron cells)were cultured and classified into control(CON),CON+ICA(10 μmol/L),model(OGD),and OGD+ICA(10 μmol/L)groups.In the OGD+ICA group,after OGD treatment,10 μmol/L ICA was added into the medium and incubated with the cells for 24 h.In the Con+ICA group,an equal dose of ICA was added.The mortality rate of neurons was detected by the cell counting kit-8(CCK8)method.The neuron apoptosis was detected by flow cytometry.Real-time PCR and Western blotting were employed to determine the mRNA and protein levels,respectively,of the neuron apoptosis markers including cysteinyl aspartate-specific proteinase 3(Caspase-3),B-cell lymphoma-2(Bcl2),and Bcl-2-associated X protein(Bax).In addition,Western blotting was employed to determine the protein level of pyruvate kinase M2(PKM2)and the expression of Caspase-3,BAX,and BCL2 in the neurons treated with the PKM2-specific agonist(TEPP-46).Results:Compared with the CON group,the OGD group showed decreased survival rate of neurons,up-regulated mRNA and protein levels of Caspase-3 and Bax,down-regulated protein level of BCL2,and up-regulated protein level of PKM2(P<0.05).Compared with the OGD group,the ICA treatment significantly increased the survival rate and decreased the apoptosis rate of neurons,and the optimal con-centration of ICA was 10 μmol/L.In addition,the ICA treatment down-regulated the mRNA and protein levels of Caspase-3 and Bax(P<0.05)and the protein level of PKM2,while up-regulating the protein level of BCL2(P<0.05).The treatment with TEPP-46 reversed the regulatory effects of ICA on the protein levels of Caspase-3 and BAX(P<0.05).Conclusion:ICA can inhibit the apoptosis and increase the survival rate of OGD neurons by regulating the expression of PKM2 to lower the levels of pro-apoptotic proteins.关键词
