摘要

Abstract

Objective:To explore the effect and mechanism of Cyclocarya paliurus on hepatic lipid metabolism in rats with diabetes mellitus and nonalcoholic fatty liver disease based on the AKt/mTOR/SREBP-1c signaling pathway.Methods:Sprague-Dawley(SD)rats were randomly divided into a normal control group,a model control group,a positive control group(10 mg/kg pioglitazone),and three Cyclocarya paliurus groups(0.45,0.90,and 1.80 mg/kg).After one month of continuous gavage,tail vein blood samples were collected to assess fasting blood sugar(FBS)levels in rats.The levels of alanine aminotransferase(ALT),aspartate aminotransferase(AST),triglyceride(TG),and total cholesterol(TC)were measured using an automatic biochemical analyzer,and the contents of malondialdehyde(MDA),glutathione(GSH),and superoxide dismutase(SOD)in liver tissue were determined using kits.Histopathological analysis of liver morphology was performed u-sing H&E staining,and hepatic lipid accumulation was observed using oil red O staining.The microstructure of liver tissue was observed via transmission electron microscopy.Additionally,mRNA expression levels of Cd36 and Atgl in rat liver were quantified using qPCR,and the protein expressions of AKT,p-AKt,mTOR,p-mTOR,SREBP-1c,LC3,and P62 were assessed by Western blot.Results:Compared with the normal control group,the model control group exhibited significantly up-regulated FBS,TG,TC,ALT,AST,MDA,and Cd36 mRNA expressions(P<0.01),increased p-AKt,p-mTOR,P62,and SREBP-1c protein expressions(P<0.01),and significantly reduced levels of LC3II/LC3I(P<0.01).In the model control group,the rat liver structure was disordered,with pronounced steatosis.Compared with the model control group,the 1.80 mg/kg Cyclocarya paliurus group displayed significantly down-regulated MDA,p-Akt,p-mTOR,P62,and SREBP-1c protein expressions,reduced Cd36 mRNA expression(P<0.01),elevated LC3II/LC3I,and significantly up-regulated Atgl mRNA expression(P<0.01).Conclusion:Cyclocarya paliurus alleviates hepatic lipid metabolism disorder and oxidative stress in rats with diabetes mellitus com-bined with nonalcoholic fatty liver disease.The underlying mechanism may involve the inhibition of AKt/mTOR/SREBP-1c signaling path-way.
