枳壳潜在燥性成分3,5,6,7,8,3',4'-七甲氧基黄酮对正常小鼠津液及胃水通道蛋白的影响研究
Effects of Potential Drying Component 3,5,6,7,8,3′,4′-heptamethoxyflavone in AURANTII FRUCTUS on Aquaporin in Stomach and Body Fluids in Normal Mice

Abstract
Objective:To study the effects of 3,5,6,7,8,3′,4′-heptamethoxyflavone(HMF),a potential drying component of AURANTII FRUC-TUS,on aquaporin(AQP)in stomach and body fluids in normal mice and to establish a new strategy for research on the scientific connotation of drug properties.Methods:Male C57BL/6 mice were randomly divided into a normal group,a 4 g/kg AURANTII FRUCTUS group,and 0.4 mg/kg,0.8 mg/kg,and 1.6 mg/kg HMF groups.The following indicators of each group were determined:24 h drinking water intake,food intake,2 h defecation rate and fecal water content,salivary secretion,small intestinal propulsion distance,and water content of gastric and sub-mandibular gland tissue.Hematoxylin-eosin(HE)staining was used to observe and score the pathological morphology of gastric tissue.The expression of AQP3,AQP5,and AQP7 extracted from gastric tissue was assessed by immunohistochemistry(IHC).Results:Compared with the normal group,the 0.8 mg/kg and 1.6 mg/kg HMF groups showed significantly increased drinking water intake(P<0.01),and the 0.8 mg/kg HMF group and the 4 g/kg AURANTII FRUCTUS group had significantly increased food intake(P<0.01).Additionally,the sali-vary secretion,2 h defecation rate,and fecal water content were significantly reduced in the 0.4 mg/kg,0.8 mg/kg,and 1.6 mg/kg HMF groups and the 4 g/kg AURANTII FRUCTUS group(P<0.01).The small intestinal propulsion distance was significantly reduced in the 0.8 mg/kg HMF group(P<0.05),while it exhibited highly significant reductions in the 0.4 mg/kg and 1.6 mg/kg HMF groups and the 4 g/kg AURANTII FRUCTUS group(P<0.01).The water content of submandibular gland tissue in all the medication groups was significant-ly reduced(P<0.05 or P<0.01),which was in the same trend of salivary secretion.Pathological damages(edema,inflammation,necrosis,etc.)in the gastric tissue were observed,with the damage scores of gastric tissue in mice of all the HMF treatment groups significantly in-creasing(P<0.01).The AQP3 expression was significantly increased in the 0.8 mg/kg HMF group(P<0.05),and the AQP5 expression in gastric tissue was significantly increased in the 4 g/kg AURANTII FRUCTUS group and the 0.8 mg/kg and 1.6 mg/kg HMF groups(P<0.01),while the AQP7 expression in gastric tissue was significantly increased in the 0.8 mg/kg and 1.6 mg/kg HMF groups(P<0.01).Conclusion:The drying property of AURANTII FRUCTUS and HMF can affect the secretion of submandibular gland and the metabolism of gastric fluid in mice,and the mechanism may be related to up-regulating the expression of AQP3,AQP5,and AQP7 in gastric tissue.关键词
