探讨葛花解酲方调控铁死亡抑制EMT干预肝细胞癌的作用及机制研究
Research on the Efficacy and Mechanism of Gehua Jiecheng(葛花解酲)Formula in Regulating Ferroptosis to Inhibit EMT in Hepatocellular Carcinoma

Abstract
Objective:To investigate the mechanism of Gehua Jiecheng(葛花解酲)Formula(GHJCF)in inhibiting epithelial-mesenchymal tran-sition(EMT)via regulating ferroptosis in the treatment of hepatocellular carcinoma based on network pharmacology,bioinformatics,molecular docking,and cell experiments.Methods:The targets of GHJCF,ferroptosis,and hepatocellular carcinoma were retrieved from the Traditional Chinese Medicine System Pharmacology Database and Analysis Platform(TCMSP),Uniport,Gene Expression Omnibus(GEO),and String.Bioinformatics tools were used to screen the differentially expressed genes.The Human Protein Atlas(THPA)was used for protein expression analysis.Clinical correlation analysis,survival and prognosis analysis,and gene set enrichment analysis(GSEA)of hepatocellular carcinoma were carried out.The core targets were predicted based on the results of the analyses above.Molecular docking was performed in Discovery Studio 2016 to screen the compounds binding to the core targets.Transwell assay and Western blotting were employed to explore the effect of GHJCF-containing serum on EMT of HepG2 cells.Results:A total of 13 targets shared by GHJCF,ferroptosis,and hepatocellular carcinoma were obtained,from which one core target was finally screened out as androgen receptor(AR).GSEA results showed that AR was closely re-lated to the cell adhesion process in EMT.The results of molecular docking showed that AR had high affinity with deoxyharringtonine,3-me-thoxy puerarin,puerarin,and genistein 8-C-glucoside.The results of Transwell assay and Western blotting showed that compared with the nor-mal control group,GHJCF-containing serum reduced the migration of HepG2 cells,up-regulated the expression levels of E-cadherin(epithelial marker)and AR(P<0.01),and down-regulated the expression levels of N-cadherin(mesenchymal marker)and glutathione peroxidase 4(GPX4,ferroptosis marker)(P<0.01)in HepG2 cells.Conclusion:GHJCF can interfere in HCC by regulating ferroptosis and inhibiting EMT via the active components acting on AR.关键词
